Melanotan II (MT-2) is a synthetic cyclic analogue of alpha-MSH (alpha-melanocyte-stimulating hormone), a peptide derived from POMC (proopiomelanocortin). It is a potent, non-selective agonist at melanocortin receptors (MC1R through MC5R), with particularly strong activity at MC1R and MC4R. This broad agonism is studied in research models in connection with both its well-characterised activity on skin pigmentation (via MC1R) and its research history around sexual-arousal pharmacology and appetite modulation (via MC4R).
The pharmacology is worth understanding in some detail. MC1R is expressed on melanocytes and regulates melanin production — in research models MT-2 is studied for activation of eumelanin synthesis, associated with darker skin pigmentation without UV exposure. MC4R is expressed in the hypothalamus and plays roles in melanocortin signalling and sexual function — in research models its activation is studied for satiety signalling and, notably, pro-erectile effects in male rodent models. MC3R has roles in energy homeostasis. MT-2 activates all of these nonselectively, which produces the compound’s range of effects in research models and also complicates research interpretation where receptor-specific effects need to be isolated.
MT-2 was developed at the University of Arizona from research aimed at characterizing a compound that would drive melanin synthesis in model systems and, in turn, was studied in the context of UV-related skin biology. The broader melanocortin agonism effects became apparent during development. It has been investigated in preclinical research on sexual-arousal pharmacology, and a related derivative (bremelanotide/PT-141) shares the same MC4R-mediated mechanism.

MT-2 is a non-selective melanocortin receptor agonist with activity at MC1R, MC3R, MC4R, and MC5R. MT-2 is the research precursor that established the MC4R-mediated arousal pharmacology that PT-141 was developed from.
